    The carcinogenic effect of various multi-walled carbon nanotubes (MWCNTs) after intraperitoneal injection in rats

    Background: Biological effects of tailor-made multi-walled carbon nanotubes (MWCNTs) without functionalization were investigated in vivo in a two-year carcinogenicity study. In the past, intraperitoneal carcinogenicity studies in rats using biopersistent granular dusts had always been negative, whereas a number of such studies with different asbestos fibers had shown tumor induction. The aim of this study was to identify possible carcinogenic effects of MWCNTs. We compared induced tumors with asbestos-induced mesotheliomas and evaluated their relevance for humans by immunohistochemical methods. Methods: A total of 500 male Wistar rats (50 per group) were treated once by intraperitoneal injection with 109 or 5 � 109 WHO carbon nanotubes of one of four different MWCNTs suspended in artificial lung medium, which was also used as negative control. Amosite asbestos (108 WHO fibers) served as positive control. Morbid rats were sacrificed and necropsy comprising all organs was performed. Histopathological classification of tumors and, additionally, immunohistochemistry were conducted for podoplanin, pan-cytokeratin, and vimentin to compare induced tumors with malignant mesotheliomas occurring in humans. Results: Treatments induced tumors in all dose groups, but incidences and times to tumor differed between groups. Most tumors were histologically and immunohistochemically classified as malignant mesotheliomas, revealing a predominantly superficial spread on the serosal surface of the abdominal cavity. Furthermore, most tumors showed invasion of peritoneal organs, especially the diaphragm. All tested MWCNT types caused mesotheliomas. We observed highest frequencies and earliest appearances after treatment with the rather straight MWCNT types A and B. In the MWCNT C groups, first appearances of morbid mesothelioma-bearing rats were only slightly later. Later during the two-year study, we found mesotheliomas also in rats treated with MWCNT D - the most curved type of nanotubes. Malignant mesotheliomas induced by intraperitoneal injection of different MWCNTs and of asbestos were histopathologically and immunohistochemically similar, also compared with mesotheliomas in man, suggesting similar pathogenesis. Conclusion: We showed a carcinogenic effect for all tested MWCNTs. Besides aspect ratio, curvature seems to be an important parameter influencing the carcinogenicity of MWCNTs

    Thanks to their extraordinary properties, carbon nanotubes reveal a promising potential for applications on the nanometre scale. When filled with metals or ferromagnets, nano-wires and magnets with a protecting carbon shell are realised. Different synthesis routes are described, such as laser ablation and chemical vapour deposition. Probes for magnetic force microscopy based on ironfilled carbon nanotubes are presented, and demonstrate a high spatial resolution, with the carbon shells at the same time providing effective wear resistance. We show also the potential of carbon nanotubes for biomedical applications, in particular their suitability as magnetic nano-heaters, drug-carrier systems or sensors for diagnostic and therapeutic usage on the cellular level.Außergewöhnliche Materialeigenschaften machen Kohlenstoffnanoröhren zu einem vielseitigen nanoskaligen Werkstoff. Füllt man sie zum Beispiel mit metallischen oder ferromagnetischen Materialien, so ergeben sich durch eine Kohlenstoffhülle geschützte „Nano- Kabel“ oder Nano-Magnete. Neben verschiedenen Syntheseverfahren wie der Laserablation und der Chemischen Gasphasenabscheidung werden grundlegende physikalische Eigenschaften sowie Anwendungen in der Messtechnik und in der Medizin vorgestellt. In der Magnetkraftmikroskopie versprechen magnetisch gefüllte Kohlenstoffnanoröhren eine hohe laterale Auflösung bei gleichzeitigem Schutz des magnetischen Messsensors durch die Außenhülle. Im Bereich der biomedizinischen Anwendungen stellen Kohlenstoffnanoröhren ein nanoskaliges Transportmedium dar, das zum Transfer von Funktionsmaterialien in einzelne Zellen, zum Beispiel für magnetische Sensorik oder für Medikamententransporte, angewendet werden kann

    Influence of vortex-vortex interaction on critical currents across low-angle grain boundaries in YBa2Cu3O7-delta thin films

    Low-angle grain boundaries with misorientation angles theta < 5 degrees in optimally doped thin films of YBCO are investigated by magnetooptical imaging. By using a numerical inversion scheme of Biot-Savart's law the critical current density across the grain boundary can be determined with a spatial resolution of about 5 micrometers. Detailed investigation of the spatially resolved flux density and current density data shows that the current density across the boundary varies with varying local flux density. Combining the corresponding flux and current pattern it is found that there exists a universal dependency of the grain boundary current on the local flux density. A change in the local flux density means a variation in the flux line-flux line distance. With this knowledge a model is developped that explains the flux-current relation by means of magnetic vortex-vortex interaction.Comment: 7 pages, 14 figure

    Rearing and foraging affects bumblebee (Bombus terrestris) gut microbiota

    Bumblebees are ecologically and economically important as pollinators of crop and wild plants, especially in temperate systems. Species, such as the buff-tailed bumblebee (Bombus terrestris), are reared commercially to pollinate high value crops. Their highly specific gut microbiota, characterised by low diversity, may affect nutrition and immunity and are likely to be important for fitness and colony health. However, little is known about how environmental factors affect bacterial community structure. We analyzed the gut microbiota from three groups of worker bumblebees (B. terrestris) from distinct colonies that varied in rearing and foraging characteristics: commercially reared with restricted foraging (RR); commercially reared with outside foraging (RF); and wild-caught workers (W). Contrary to previous studies, which indicate that bacterial communities are highly conserved across workers, we found that RF individuals had an intermediate community structure compared to RR and W types. Further, this was shaped by differences in the abundances of common OTUs and the diversity of rare OTUs present which we propose results from an increase in the variety of carbohydrates obtained through foraging

    Upper limits on the strength of periodic gravitational waves from PSR J1939+2134

    The first science run of the LIGO and GEO gravitational wave detectors presented the opportunity to test methods of searching for gravitational waves from known pulsars. Here we present new direct upper limits on the strength of waves from the pulsar PSR J1939+2134 using two independent analysis methods, one in the frequency domain using frequentist statistics and one in the time domain using Bayesian inference.     Improving the sensitivity to gravitational-wave sources by modifying the input-output optics of advanced interferometers

    We study frequency dependent (FD) input-output schemes for signal-recycling interferometers, the baseline design of Advanced LIGO and the current configuration of GEO 600. Complementary to a recent proposal by Harms et al. to use FD input squeezing and ordinary homodyne detection, we explore a scheme which uses ordinary squeezed vacuum, but FD readout. Both schemes, which are sub-optimal among all possible input-output schemes, provide a global noise suppression by the power squeeze factor, while being realizable by using detuned Fabry-Perot cavities as input/output filters. At high frequencies, the two schemes are shown to be equivalent, while at low frequencies our scheme gives better performance than that of Harms et al., and is nearly fully optimal. We then study the sensitivity improvement achievable by these schemes in Advanced LIGO era (with 30-m filter cavities and current estimates of filter-mirror losses and thermal noise), for neutron star binary inspirals, and for narrowband GW sources such as low-mass X-ray binaries and known radio pulsars. Optical losses are shown to be a major obstacle for the actual implementation of these techniques in Advanced LIGO. On time scales of third-generation interferometers, like EURO/LIGO-III (~2012), with kilometer-scale filter cavities, a signal-recycling interferometer with the FD readout scheme explored in this paper can have performances comparable to existing proposals.     CDPKs CPK6 and CPK3 Function in ABA Regulation of Guard Cell S-Type Anion- and Ca(2+)- Permeable Channels and Stomatal Closure

    Abscisic acid (ABA) signal transduction has been proposed to utilize cytosolic Ca(2+) in guard cell ion channel regulation. However, genetic mutants in Ca(2+) sensors that impair guard cell or plant ion channel signaling responses have not been identified, and whether Ca(2+)-independent ABA signaling mechanisms suffice for a full response remains unclear. Calcium-dependent protein kinases (CDPKs) have been proposed to contribute to central signal transduction responses in plants. However, no Arabidopsis CDPK gene disruption mutant phenotype has been reported to date, likely due to overlapping redundancies in CDPKs. Two Arabidopsis guard cell–expressed CDPK genes, CPK3 and CPK6, showed gene disruption phenotypes. ABA and Ca(2+) activation of slow-type anion channels and, interestingly, ABA activation of plasma membrane Ca(2+)-permeable channels were impaired in independent alleles of single and double cpk3cpk6 mutant guard cells. Furthermore, ABA- and Ca(2+)-induced stomatal closing were partially impaired in these cpk3cpk6 mutant alleles. However, rapid-type anion channel current activity was not affected, consistent with the partial stomatal closing response in double mutants via a proposed branched signaling network. Imposed Ca(2+) oscillation experiments revealed that Ca(2+)-reactive stomatal closure was reduced in CDPK double mutant plants. However, long-lasting Ca(2+)-programmed stomatal closure was not impaired, providing genetic evidence for a functional separation of these two modes of Ca(2+)-induced stomatal closing. Our findings show important functions of the CPK6 and CPK3 CDPKs in guard cell ion channel regulation and provide genetic evidence for calcium sensors that transduce stomatal ABA signaling

    Searching for a Stochastic Background of Gravitational Waves with LIGO

    The Laser Interferometer Gravitational-wave Observatory (LIGO) has performed the fourth science run, S4, with significantly improved interferometer sensitivities with respect to previous runs. Using data acquired during this science run, we place a limit on the amplitude of a stochastic background of gravitational waves. For a frequency independent spectrum, the new limit is ΩGW<6.5×105\Omega_{\rm GW} < 6.5 \times 10^{-5}. This is currently the most sensitive result in the frequency range 51-150 Hz, with a factor of 13 improvement over the previous LIGO result. We discuss complementarity of the new result with other constraints on a stochastic background of gravitational waves, and we investigate implications of the new result for different models of this background.Comment: 37 pages, 16 figure
